Assay of Tissue Phosphoglucomutase

ثبت نشده
چکیده

Although several values for tissue phosphoglucomutase activites are recorded in the literature, such determinations have been performed without the addition of the coenzyme, a-glucose 1,6-diphosphate (l-3), and occasionally without explicit designation of the optimal concentrations of either or both of the known activators, Mg++ and amino acid (2, 3). Although some of these latter factors have been previously considered (4), the present paper is concerned with the extent to which added coenzyme influences the mutase activity of tissue homogenates and the conditions under which coenzyme and the other activating compounds give maximal phosphoglucomutase activity and hence yield a precise method of assay for this enzyme in tissue.

Studies on Phosphoglucomutase and Phosphofructokinase from Brain

It has recently been established that the activity of crystalline muscle phosphoglucomutase can be greatly stimulated by preincubation of the enzyme with a Mg++-imidazole complex. This observation has aroused interest in the physiological significance of such a system i n the possible cellular control of phosphoglucomutase activity. The present study constitutes, i n part, an investigation of t...

Functional diversity of the phosphoglucomutase superfamily: structural implications.

Three-dimensional structural models of three members of the phosphoglucomutase (PGM) superfamily, parafusin, phosphoglucomutase-related protein and sarcoplasmic reticulum phosphoglucomutase, were constructed by homology modeling based on the known crystal structure of rabbit muscle phosphoglucomutase. Parafusin, phosphoglucomutase-related protein and sarcoplasmic reticulum phosphoglucomutase ea...

Evidence for a light dependent increase of phosphoglucomutase activity in isolated, intact spinach chloroplasts.

Phosphoglucomutase (PGM) activity was measured in spinach (Spinacia oleracea L.) chloroplasts. Initial enzyme activity in a chloroplast lysate was 5 to 10% of total activity measured with 1 micromolar glucose 1,6-bisphosphate (Glc 1,6-P(2)) in the assay. Initial PGM activity increased 2- to 3-fold when chloroplasts were illuminated for 10 minutes prior to enzyme measurement and then decreased s...

The inhibition of enzymes by beryllium.

1. The action of beryllium on the following enzymes has been examined: alkaline phosphatase (Escherichia coli and kidney), acid phosphatase, phosphoprotein phosphatase, apyrase (potato), adenosine triphosphatase (liver nuclei, liver mitochondria, brain microsomes), glucose 6-phosphatase, polysaccharide phosphorylases a and b, phosphoglucomutase, hexokinase, phosphoglyceromutase, ribonuclease, A...

Genetic control of phosphoglucomutase variants in Saccharomyces cerevisiae.

The three electrophoretic variants of phosphoglucomutase in Saccharomyces cerevisiae breeding stocks are produced by two unlinked genes, pgm-1 and pgm-2; pgm-1 contains two known alleles, pgm-1a and pgm-1b, each of which specifies a minor phosphoglucomutase component, and pgm-2 specifies the major phosphoglucomutase component.
